Description:
Wiskott-Aldrich syndrome protein (WASP) is a major cytoskeletal regulator expressed in hematopoietic cells. Mutations in the WASP gene result in an immune deficiency with thrombocytopenia. A 500 bp fragment from the proximal WASP gene promoter (extending from bases -1 to -499 from the transcriptional start site) was shown to direct hematopoietic-specific expression of a transgene [1]. This fragment contains four potential binding sites for hematopoietic transcription factors (two for ets-1, one for c-Rel, and one for PU.1). Transduction of WASP-deficient cells with a lentiviral expressing the WASP gene under the control of the proximal WASP gene promoter resulted in physiological levels of WASP expression [2]. The WASP expression levels were 135-350 times lower in non-hematopoietic cells.
