Description:
Endothelin-1 (ET-1) is a peptide synthesized by endothelial cells both in culture and in vivo. ET-1 induces contraction of smooth muscle cells and stimulates growth in a variety of mesenchymal cell types. We cloned and sequenced the human ET-1gene. The objective of the work reported here was to determine which cis-acting sequence in the ET-1 promoter region are important for transcription of the gene. Using delection mutants of the ET-1 promoter, promoter reconatitution experiments, and enhanser trap experments, we have identified a 119-base pair sequence containing two regions necessary for ET-1 promoter function. This sequence allows expression of a heterologous promoter in endothelial cells.
